Pipette tips are an indispensable tool in the liquid transfer process, and their cleanliness and sterility play a decisive role in the success of the experiment. So, why do pipette tips need to be sterilized? How to carry out sterilization effectively? 1. Alcohol soaking method: Pour 70% to 75% alcohol into a small container. Completely immerse the pipette tip in the alcohol and leave it there for a few minutes to ensure that the alcohol fully contacts and kills the bacteria. After sterilization, wait for the alcohol to evaporate completely before use to avoid alcohol residue affecting the experimental results. 2. High temperature and high pressure sterilization method: Place the tip into a test tube or container that is resistant to high temperature and high pressure, add an appropriate amount of deionized water, be careful not to fill it up, and leave space. Cover the mouth of the test tube with sterile gauze or cotton swabs and place it in a high-temperature and high-pressure sterilizer. Follow the instructions for use of the sterilizer and generally maintain a high temperature and high pressure environment for 20 to 30 minutes. 3. Moist heat sterilization method: Place the tip in a container filled with water, heat to boiling and maintain for a period of time, such as 121°C for 15 to 20 minutes. This method is suitable for tips that can withstand high temperatures. To avoid moisture problems, wrap the tip box in newspaper or place it in an insulated box to dry after disinfection. Newspaper absorbs water and most importantly prevents re-contamination. 4. Ultraviolet sterilization method: Place the pipette tip into a UV sterilizer and ensure that the UV lamp can illuminate all surfaces of the tip. Turn on the ultraviolet lamp and irradiate according to the set time of the sterilizer. The general processing time is 15 to 30 minutes. After sterilization, rinse the tip with sterile deionized water, then dry or blow dry it before use. 5. Chemical reagent sterilization method: use chemical reagents such as hydrogen peroxide and acetic acid for treatment. Add an appropriate amount of chemical reagent into the sterile test tube, place the pipette tip in the test tube, and cover the mouth of the test tube. Operate according to the temperature and time conditions approved by the drug. After sterilization, take out the test tube and wash it with deionized water to remove any residual drugs on the surface. 6. Enzyme detergent sterilization method: Immerse the pipette tip in the enzyme detergent solution and keep it for a certain period of time. The enzyme detergent can remove bacteria and organic matter on the surface of the tip. After sterilization, rinse the tip with sterile deionized water, then dry or blow dry it before use.
When selecting a sterilization method, comprehensive considerations need to be made based on the material of the pipette tip, experimental requirements, and laboratory conditions. At the same time, in order to ensure the sterilization effect, it is necessary to operate in accordance with the requirements of the sterilization method and pay attention to preventing recontamination.
