1. High-temperature and high-pressure steam sterilization method 1. Operation steps: (1) Place the pipette tip box and the tips inside it into a high-pressure steam sterilization pot. (2) Set the sterilization temperature to 121°C, the pressure to 102.9kPa (or 1.05kg/cm²), and the sterilization time to 20 to 30 minutes. (3) After sterilization is completed, wait for the high-pressure steam sterilizer to cool down naturally or perform rapid cooling according to the instructions. (4) Open the sterilization pot, use sterile tools to take out the pipette tip box, and place it in a sterile environment for later use. 2. Precautions: (1) Ensure that the pipette tip box and the tips inside it can withstand high temperature and pressure. (2) During the sterilization process, pay close attention to the status of the sterilization pot to ensure that the sterilization conditions are met. (3) After sterilization, take out the pipette tip box promptly to avoid deformation or damage caused by long-term exposure to high temperature environment.
2. Chemical disinfection method 1. Operation steps: (1) Choose appropriate chemical disinfectants, such as 75% ethanol, hydrogen peroxide, etc. (2) Soak the pipette tip box and the tips inside it in the disinfectant to ensure complete immersion. (3) The soaking time depends on the type and concentration of the disinfectant, generally ranging from several minutes to tens of minutes. (4) After soaking, use sterile water to rinse the pipette tip box and the tips inside to remove residual disinfectant. (5) Place the pipette tip box in a sterile environment to dry or use sterile paper to dry it. 2. Precautions: (1) Choose a suitable disinfectant to ensure that it is non-corrosive to the pipette tip box and the tips inside it. (3) The residual disinfectant must be completely removed during the flushing process to avoid interference with subsequent experiments.
3. Ultraviolet disinfection method 1. Operation steps: (1) Place the pipette tip box and the tips inside it under ultraviolet light. (2) Turn on the ultraviolet lamp. The irradiation time depends on the intensity of the ultraviolet rays and the resistance of the bacteria to ultraviolet rays. It is generally 15 to 30 minutes. (3) After the irradiation is completed, turn off the UV lamp, use sterile tools to remove the pipette tip box, and place it in a sterile environment for later use. 2. Precautions: (1) The ultraviolet disinfection method is suitable for disinfecting the surface of the pipette tip box, and the disinfection effect on the internal tips may be limited. (2) Ultraviolet rays are harmful to the human body. Protective glasses and gloves must be worn during operation. (3) The irradiation intensity and irradiation time of the ultraviolet sterilizer need to be adjusted according to specific conditions to ensure the disinfection effect.
4. Other precautions 1. Before performing any disinfection operation, be sure to read the instructions for the pipette tip box and its internal tips to understand its material, tolerance and disinfection requirements. 2. During the disinfection process, aseptic operation principles must be followed to avoid cross-contamination. 3. After the disinfection is completed, the pipette tip box and the tips inside should be inspected for sterility to ensure their sterility.
